A new systematic review found that cannabidiol (CBD) produces antidepressant-like effects, potentially through multiple biological mechanisms involving neuroplasticity, inflammation and neurotransmitter signaling.
The study, published August 7 in the Pará Research Medical Journal, was conducted by researchers from Universidade Anhembi Morumbi in Brazil. Researchers reviewed experimental studies published during the previous five years examining CBD and related cannabinoids for antidepressant-like effects.
Using the PRISMA systematic-review protocol, researchers identified seven experimental studies that met their inclusion criteria. Across the studies, CBD was associated with reduced immobility in the forced swim test and increased preference for sucrose, two measures commonly used in animal models to assess antidepressant-like activity and depression-related behavior.
The review found that CBD’s effects appeared to involve several molecular pathways rather than a single mechanism. These included activation of AMPA receptors and modulation of serotonin and norepinephrine transmission, both of which play important roles in mood regulation.
CBD was also associated with reduced expression of the inflammatory markers NF-κB and IL-6 and regulation of the kynurenine pathway. Researchers additionally found evidence that CBD influences neurogenesis in the hippocampus, a brain region involved in memory and mood that has been extensively studied in relation to depression.
Other findings pointed to anti-inflammatory and antiapoptotic effects involving BDNF/TRPC6 signaling, a pathway associated with neuronal survival and neuroplasticity. The reviewed research also included evidence that chronic synthetic CBD administration can alter serotonin and norepinephrine levels in the hippocampus.
Researchers concluded that the findings demonstrate a multifaceted antidepressant potential for CBD and related compounds and suggest they could eventually have a role as adjunctive or alternative treatments for mood disorders.
However, the evidence reviewed was preclinical rather than clinical, meaning the findings do not establish that CBD is an effective treatment for depression in humans. Researchers said randomized human clinical trials are needed to determine its efficacy, safety and appropriate treatment parameters.
