Study Finds Single Dose of CBD Restored Blood-Brain Barrier Integrity and Reduced Depression-Like Behaviors in Animal Model

A new study found that a single dose of cannabidiol (CBD) restored key measures of blood-brain barrier integrity, reduced neuroinflammation and improved depression-like behaviors in mice.
The research, published in CNS Neuroscience & Therapeutics, examined whether CBD could counter blood-brain barrier dysfunction associated with depression and explored the mechanisms responsible for the effects. Researchers from the Academy of Military Medical Sciences and Chinese PLA Medical School conducted the study.
Researchers induced a chronic inflammation-associated depression model in male mice using lipopolysaccharide (LPS) for seven days. The animals then received a single 30 mg/kg dose of CBD, followed by behavioral testing and analyses of blood-brain barrier function, astrocyte activity and inflammatory markers.
Mice exposed to LPS displayed reduced exploratory behavior and increased depression-like behaviors. They also showed several signs of blood-brain barrier impairment, including reduced expression of proteins responsible for maintaining tight junctions between endothelial cells, abnormal astrocyte activation and elevated neuroinflammation.
CBD significantly improved the behavioral measures while reversing multiple signs of blood-brain barrier damage. Researchers found that CBD restored expression of the tight junction proteins ZO-1 and occludin and reduced abnormal barrier permeability. Experiments using cultured human brain endothelial cells produced similar results, with CBD increasing electrical resistance across the barrier and reducing permeability following inflammatory damage.
The study also identified what researchers described as a potentially important mechanism involving “memory astrocytes,” a subset of brain cells that can develop an enhanced inflammatory response after previous exposure to inflammation.
CBD suppressed the accumulation of ACLY+p300+ memory astrocytes and reduced their heightened inflammatory responses. The researchers said this may decrease secondary neuroinflammation, protect the blood-brain barrier and contribute to improvements in depression-like behaviors.
The researchers said their findings suggest that CBD’s effects may involve both stabilization of the blood-brain barrier and regulation of inflammatory processes involving astrocytes.
“The present study demonstrates that a single dose” of CBD produced protective effects on the blood-brain barrier while alleviating depression-like behaviors, the researchers wrote, describing CBD as a promising candidate for further investigation.
The authors cautioned that the research was preclinical and primarily involved mice and laboratory cell models. They also noted that the experiment examined acute, short-term CBD treatment, meaning longer-term studies are needed to determine efficacy, safety and tolerability. Further research is also needed to establish whether the proposed memory-astrocyte pathway is directly responsible for CBD’s effects.
The researchers concluded that CBD merits further investigation as a potential approach for reducing blood-brain barrier damage and depressive symptoms through its effects on pathological astrocyte activity.
